Whatnow, thank you for this information about the daily dosing. That would be highly encouraging if more dosing meant even more AD benefit. Is the AD benefit from Viagra thought to also include an amyloid component, from what I remember reading it was reported to possibly work more through tau? It is quite exciting to realize that these repurposing might uncover mechanisms of action that had been entirely overlooked.

My suspicion is that now that effective AD medicines are approaching shoreline that other treatments far on the back
shelf might be dusted off. Basically, it is now or never. AD is in the process of being cured. Those who want to be part of the cure need to get on board the train (the gravy train) now before it leaves the station.

I found the article about increasing frequency of viagra use being associated with greater benefit. It was yet another retrospective study on heart disease, not Alzheimer's. Also, the articles I've read on possible benefit for Alzheimer's indicated it they were looking for drugs that analysis indicated might have an effect on both Tau and Amyloid. This was what led them to look at Viagra.

Are we sure the decreased AD isn't because would be AD patients already died of heart attacks? ED is linked to coronary issues.

From a research group at Mt Sinai using a large data-based analysis of over 50k individuals:

"The propensity score stratified univariate analysis and the Cox regression model showed that sildenafil
use was significantly associated with a 60% risk reduction of developing
Alzheimer's disease (HR=0.40; 95%CI:0.38-0.44; P<.0001) compared to the
cohort of individuals who did not take sildenafil. Sex-stratified analyses revealed
that sildenafil was related to a lower risk of Alzheimer's disease in subgroups of
both males and females. Our findings demonstrated a significant association
between sildenafil use and a lower risk of Alzheimer's disease."

This is a study on humans, not mice. Sixty percent reduction in developing Alzheimer's seems almost too good to be true. The paper (attached) outlines how the data was analyzed.

Would anyone like to hazard a guess as whether these effects would also be seen in tadalafil and vardenafil?

Maybe it's more about increase in physical activity than Viagra itself
